Research in the Meistrich laboratory focusing on regulation of differentiation of spermatogonial stem cells. The goal of the research is to elucidate the molecular and cellular mechanisms underlying our discovery that treatment of rats with radiation, chemotherapy drugs, or some environmental toxicants induces a complete block in spermatogonial differentiation, but that this block is reversed by suppressing testosterone. This provides a direct approach to studying the role of hormonally regulated paracrine factors produced by somatic cells on spermatogonial differentiation. The techniques used to investigate these questions include molecular studies of the endocrine regulation of gene expression in vivo and in isolated androgen-responsive somatic cell subpopulations and in tissue culture where we can also assess the progression of differentiation, spermatogonial stem cell and somatic cell transplantation to restore damaged function, and modeling these effects in genetically engineered rodents.
