The Life Sciences Division has an opportunity for a Biologist Postdoctoral Fellow to explore the regulation of alternative pre-mRNA splicing during erythroid differentiation. Experiments will be designed to explore molecular mechanisms by which splicing factor Fox2 activates the splicing of protein 4.1R exon 16 in erythroblasts. Fox2 is a key splicing factor that appears to play a critical role in regulation of many tissue-specific splicing events, and exon 16 provides a very useful model system to study molecular mechanisms of Fox2 activity. A variety of erythroid and nonerythroid cell models will be used to explore the interaction between antagonistic hnRNP A1 and Fox2 splicing factors, and to identify other splicing factors that bind to the highly conserved intron sequence flanking the Fox sites. The role of a highly conserved distal intron regulatory element will also be investigated. Finally, the project will study the importance of Fox2 regulation in vivo using mouse knockin technology to mutate the 4.1R intron 16 Fox2 binding sites. The goal of these studies is to develop a more general understanding of the factors regulating a physiologically important splicing event and to understand the activity of a crucial alternative splicing factor.

As a Postdoc, you will:
• Design and perform experiments utilizing molecular biology, biochemistry, and potentially microarrays to understand the regulation of alternative pre-mRNA splicing in differentiating erythroid cells.
• Prepare results for publication in scientific journals and presentations at seminars.
• Publish research materials in leading journals.
• Maintain a clear record of research progress and data collected.

Desired Traits
• Ph.D. within the last 2 years in cell and molecular biology or related field.
• Demonstrated ability to manipulate and utilize recombinant DNA.
• Demonstrated skill utilizing molecular genetic and cell biology tools for analyzing gene function.
• Ability to work with guidance of principal investigator and collaborators.
• Ability to conduct experiments independently, maintain appropriate records and manage data.
• Excellent verbal and written communication skills.
• Familiarity with generation and analysis of gene expression data.
• Experience in culture of human cells.

This is a full time, one-year term appointment with the possibility of renewal contingent upon continued funding and satisfactory performance.
